The Science

Longevity & Skin Health

Researchers measured women's skin during a 5-day fast. Here is what the instruments recorded.

Most skincare is judged by how it looks in the mirror. A clinical study took a different approach — it put skin under instruments that measure hydration and roughness directly, before and after the Prolon 5-Day Fasting Mimicking Diet. The numbers tell a story about where skin health actually begins at the cellular level.

For decades, the skincare conversation has lived almost entirely at the surface. Serums, peels, lasers — every category is built on the same premise: treat the outermost layers, and the rest will follow. It is a reasonable place to start. It is also the only layer most products can reach.

A line of research out of the longevity field has been working from the opposite direction. The question it asks is not what can we apply to skin, but what happens to skin when you change what the cells underneath it are doing. And the most striking part of that research is not a marketing claim. It is a set of measurements — taken by instruments, verified by an independent observer, recorded before and after a five-day nutrition program.

The clinical research at the center of this article measured skin hydration, roughness, and overall skin quality before and after the Prolon 5-Day Fasting Mimicking Diet — with the skin results recorded after three consecutive monthly cycles of the program. 1 The 5-Day program's claim is specific: that five days of precision nutrition can prompt cellular renewal deep enough to register in the skin.

What the study measured

The clinical study did not rely on people's impressions of their own skin. It used three kinds of evidence, layered on top of each other so that no single soft measure carried the conclusion:

Instrument readings. Machines measured skin hydration and skin roughness directly — the same category of objective tooling a dermatology lab would use, not a subjective glow rating.2 Independent verification. An observer who was not running the program assessed skin quality, so the scoring did not depend on the people hoping for a result. A follow-up window. Skin was not only checked at the finish line. Researchers came back well after the program ended to see whether anything held.

The headline number: hydration

The clearest, most-reported finding from the research is skin hydration. After three consecutive monthly cycles of the 5-Day program, measured skin hydration rose by 25%.2 Hydration is one of the most direct readouts of skin-barrier health, and it is the variable most topical products spend the most effort trying to move.

What surprised the researchers was not only that hydration rose, but that it stayed elevated. When they came back at day 71 — weeks after participants had finished — measured hydration was still significantly higher than where it started.1 A topical effect can fade soon after you apply it. A change that persists after the input stops points to something happening below the surface.

Hydration and surface roughness were measured by instrument — objective tooling rather than self-reported impressions.

Alongside hydration, the study reported improvement in skin roughness, fine-line appearance, and overall skin quality. Roughness and quality were the measures the instruments and the independent observer were built to catch. The direction of every measure ran the same way: toward healthier-looking skin.1

Why fasting can help improve skin

Most people associate fasting with the opposite of healthy skin — water fasts and juice cleanses leave skin looking drawn, because depriving the body of nutrients forces it to pull resources away from less essential functions, and skin is near the front of that line.

The 5-Day Program is built specifically to avoid that. Not a water fast, and not a cleanse, it provides five days of precision nutrition designed to keep the body in a fasting state while still providing food.3 The skin gets nourished, while the body still registers it as a fast.

During those days, the body shifts into a state of autophagy, in which older, damaged cell components are cleared out to make room for newer, younger functioning ones. That cellular housekeeping is the mechanism the skin study was built to evaluate — and the most plausible explanation for why a hydration gain would still be measurable 71 days later.

Hydration and surface roughness were measured by instrument — objective tooling rather than self-reported impressions.

The institution behind the research

The reason this study reads differently from the average skincare claim is where it comes from. The Fasting Mimicking Diet was developed by Dr. Valter Longo and his team at the USC Longevity Institute, built over 25+ years of research.3

Developed at the USC Longevity Institute. The Fasting Mimicking Diet originated in Dr. Valter Longo's longevity research and is the program used in the skin study.3

That research background matters for one reason: a topical product does not need a longevity lab behind it to claim a glow. A nutrition program that says it changes skin from the inside does.

How to read these results honestly

A few caveats, in the spirit of the science. The headline hydration figure is the one with a hard number behind it; the roughness, fine-line, and observer-quality improvements were reported as directional findings, which is why this article shows them as improvements rather than inventing exact figures. Results in any nutrition study reflect averages across participants, not a guarantee for any one person. And the program supports skin health and appearance — it is not a treatment for any skin condition.

What the study does support is narrow and worth stating plainly: across the measured group, a five-day precision-nutrition program produced an objectively measured rise in skin hydration that persisted weeks later, alongside directional improvements in roughness, fine lines, and overall skin quality — verified by instruments and an independent observer.1,2For an audience trained to be skeptical of glow claims, that is a meaningfully different kind of evidence.
