Glossary of Terms — understanding the benefits.
General Terms
1. Autophagy
Autophagy
The cells’ natural repair process in which they clean out, break down, and recycle their own components to improve function and efficiency, generate energy, or build new cellular structures resulting in cellular renewal or rejuvenation.
.
Autophagy plays a crucial role in cellular quality control, adaptation to stress, and the prevention of various diseases, including cancer, neurodegenerative disorders, and metabolic diseases. Autophagy is a process that typically activates on day 3 of a prolonged fast such as Prolon’s 5-Day Fasting Mimicking Diet.
2. Biological Age Score
Biological Age Score
The estimated age of our cells and bodily tissues, which does not always match our chronological age (how many years we have been alive). Determined by factors such as genetics, lifestyle, nutrition, and overall health status, biological ages core can be influenced by choices and changes in a person’s diet and lifestyle. Prolon’s 5-Day FMD has shown to potentially reduce biological age score by anaverage of 2.5 years after three consecutive cycles.
3. Healthspan
Healthspan
A term used to describe the healthy years of a person’s life; it is not just about adding years to your life but ensuring those years are healthy, vibrant, and full of vitality. In western cultures, while lifespans are increasing, people are getting sicker and sicker at younger ages. The focus needs to shift to emphasize our healthspans, not just our lifespan, so that we can be healthy as we live these longer lives. L-Nutra and Prolon’s products are designed to optimize healthspan.
4. Lifespan
Lifespan
The time a person lives, from birth to death.
5. Longevity Sites
Longevity sites:
Regions in the world with the highest populations of centenarians (people who live healthily into their hundreds).
6. Longevity Diet
The Longevity Diet
The Longevity Diet was developed by Dr. Valter Longo, based on years of research studying the eating habits of those in regions with the largest populations living healthily into their 100s. The diet emphasizes the consumption of fresh foods from plant-based sources, periodic prolonged fasting (Prolon’s 5-Day FMD), and maintaining a nutrient composition of 55-65% carbohydrates, 30-35% fats, and 10-11% protein, with meals consumed within a 12-hour TRE circadian eating window. It recommends a mostly vegan diet that is low in sugar, high in fiber and healthy fats, avoiding processed foods, and limiting alcohol intake.
7. Nutrition Technology
Nutrition Technology
The specific nutritional formulation Prolon’s products use to provide nourishment to the body while keeping cells in a fasting mode. This formulation is based on 25 years of research and development, and is used to develop different products formulations targeted to different solutions.
Did you know?
Prolon’s Fasting Bars and Fasting Shake were formulated to support intermittent fasting and a time restricted eating schedule by providing nourishment without triggering the body’s nutrient sensing pathways, keeping it in a fasting, fat-burning state. Either can be used daily as a meal or snack replacement during the fasting window and to help elongate your fast. The 1-Day Reset can be used to incorporate 24-hour periodic fasting, or within a longer intermittent program.
Fasting & Nutrition Technology
1. Prolonged or Periodic Fasting
Prolonged or periodic fasting
A fast that is longer than two days and can activate autophagy. Prolonged fasting also offers deeper metabolic reset and more weight care support than a shorter fast, providing potentially greater longevity benefits than shorter fasts.
.
2. Intermittent Fasting (IF)
Intermittent Fasting (IF)
Up to 2 days of fasting which can support metabolic health and weight loss or management but is not long enough to trigger autophagy or cellular fasting. Examples of fasting/feeding windows in Intermittent Fasting may be 12:12 (12 hours on/12 off, following the circadian eating pattern that L-Nutra recommends), 16:8, or 5:2 (five days eating/ two days off).
3. Time Restricted Feeding/Eating (TRE)
Time Restricted Feeding/Eating (TRE)
The most common form of Intermittent Fasting, TRE refers to the hours of the day that you are eating, versus the hours of the day that you are not. Condensing eating times hosts a wide variety of benefits including improvements in energy levels, weight management/loss, sleep, insulin sensitivity, and more. L-Nutra recommends following a TRE circadian eating pattern known as a 12:12 if weight loss is not your goal. The 12:12 pattern limits your daily eating window from sunrise to sundown, aligning with your body's internal clock, known as circadian rhythms.
4. Traditional Fasting
Traditional Fasting (water fasting):
A traditional water fast requires abstinence from all food and anything but water. While it has many benefits such as weight loss, fat-burning through ketosis, and cellular rejuvenation due to autophagy (when followed for more than two days), there are some risks involved, including loss of muscle, nutritional deficiencies, health risks for those with certain medical conditions, or slowing metabolism when done for too long or too frequently.
5. Physiological Fasting
Physiological Fasting (ketosis):
The phase the body typically goes through around day two of a fast, when it switches from burning external glucose from food as energy to burning internal fatty acids and ketones (typically found on the body as stored fat) as its main source of fuel.
.
Prolon’s 1-Day Reset Fast transitions the body into a fasting state and activates ketone pathways through precise nutrition formulations.
6. Nutrition Technology

7. Fasting Mimicking Diet (FMD)
Fasting Mimicking Diet (FMD), cellular fasting, fasting with food:
Fasting Mimicking Diet (FMD) was developed after 25 years and over $48 million at the Longevity Institute of the University of Southern California along with over 18 other universities and medical research centers. Its nutrition technology allows for precise food intake that nourishes the body without triggering the food sensing pathways, thereby mimicking the effects of fasting on the body. FMDs stimulate healthy stress response pathways by inducing a mild state of cellular stress, triggering adaptive responses promoting cellular repair (autophagy), rejuvenation, and metabolic health. Fasting with food enables the body to undergo a prolonged period of safe and nourished fasting without having to face some of the potential challenges that come with a water-only fast. Those Issues include adherence and possible health risks such as dehydration, muscle loss, the worsening of present health conditions, and nutrient deficiency, are examples. Prolon 5-Day is L-Nutra’s patented FMD and is the only commercially available FMD on the market.
8. Prolon 5-Day Fasting Mimicking Diet (FMD)
Prolon 5-Day Fasting Mimicking Diet (FMD)
The first and only patented nutrition program that provides the body with precise formulations that have been clinically tested to mimic and enhance the benefits of fasting, and supports cellular rejuvenation, improved metabolic health, and enhanced autophagy. L-Nutra, the makers of 5-Day Prolon, recommend initially following the 5-Day program for three consecutive months(5 days on, 25 days off, repeated three months in a row), and then three times a year after that to receive the full scope of benefits, including potential reduction of biological age score by 2.5 years (and potentially 11yrs over 20 years), weight loss with muscle protection, and metabolic marker reset. As with all nutrition-based programs, it is recommended for those with medical conditions to consult a healthcare professional before beginning.

FUNCTION AND PATHWAYS
1. Ketones and Ketosis
Ketones and Ketosis
Ketones are the acids produced by the body when it breaks down fat for fuel instead of glucose during the process of ketosis. This can happen when the body doesn't have enough glucose stored, such as during fasting (such as during the 5-Day FMD), or when following a low-carb diet. For those who are practicing Intermittent Fasting, Prolon’s Fasting Bars and Fasting Shakes extend this period of fast burning by staying under nutrient sensing pathways, so the body still thinks it is fasting.
2. Nutrient-sensing pathways (NSPs)
Nutrient-sensing pathways (NSPs)
Nutrient sensing pathways recognize the presence of nutrients & help the body make decisions about how to use and distribute consumed nutrients effectively. For example, when these pathways detect certain nutrients, they send signals to various parts of the body, instructing them on what to do, such as energy utilization, metabolism regulation, and cell growth and repair. During fasting, these sensors are not triggered by nutrients, and by their suppression the body reacts by conserving energy & minimizing any growth activities. Prolon was formulated to avoid triggering three key nutrient sensing pathways: IGF-1 (a protein pathway), PKA (carbohydrate pathway), and mTor (amino acids pathway). By formulating foods that do not trigger these nutrient-sensing pathways, L-Nutra has made it possible for the body to stay in a fasting state, even while eating.
Prolon’s 5-Day FMD, 1-Day Reset, Fasting Bars, and Fasting Shake were all formulated to stay below these NSPs.
3. IGF-1
IGF-1 (insulin-like growth factor 1)
Is a growth hormone produced primarily in the liver that is crucial for growth, tissue and muscle maintenance, and overall body function. It is vital during adolescence and childhood and throughout life for muscle and organ maintenance but, when triggered excessively with overconsumption of protein, can accelerate aging. Conversely, inadequate protein intake can lead to low IGF-1 levels, affecting muscle mass and aging. Finding the right balance of IGF-1 is essential for optimal health, as high levels have been shown to increase risks of diseases like cancer, metabolic, and cardiovascular issues. Animal-based proteins such as meat, poultry, dairy, or whey, stimulate IGF-1 more so than plant based sources.
Prolon’s L-Protein is the first of its kind plant-based protein powder formulated to provide a balance between adequate protein intake while moderating IGF-1 levels for graceful aging.
4. mTOR
mTOR (mechanistic target of rapamycin)
Regulates cellular processes like growth, metabolism, and protein synthesis. When dysregulated, mTOR signaling is associated with diseases like cancer, metabolic disorders, and neurodegenerative diseases. Foods that are high in protein, particularly those containing the amino acid leucine such as meat, fish, dairy, eggs, and soy products, can trigger mTOR signaling. High-glycemic carbohydrates such as refined grains and sugary foods and drinks may also trigger it.
5. PKA
PKA (Protein kinase A)
Is an enzyme that modifies the activity of target proteins by adding a phosphate group to them. It regulates various physiological processes such as metabolism, cell growth, muscle contraction, and nerve signaling, and triggers different cellular responses like changes in gene expression, metabolism, and cell growth. Imbalances in PKA signaling are linked to diseases like cancer, diabetes, and neurological disorders. Certain nutrients and compounds like sugars, carbohydrates, and caffeine can indirectly trigger PKA.
6. Stress Resistance Pathway
Stress Resistance Pathway
Stress resistance pathways are activated when the body encounters stress - physical, emotional, or environmental - to help it maintain balance and adapt to and overcome the triggers. These pathways involve various biological processes that are crucial for cell survival and function under adverse conditions.Fasting Mimicking Diets positively stimulate stress response pathways by inducing a mild state of cellular stress, triggering adaptive responses promoting cellular repair(autophagy), rejuvenation, and metabolic health.
